SCH-5472. is a stimulant drug developed by Schering-Plough in the 1950s. Synthesis Potassium amide was made from the reaction of potassium metal with liquid ammonia. The coupling between the carbanion created from diphenylmethane 01-81-5(1) and ethyl 2-furoate 14-99-3 335-40-6(2) gives 2-furyl benzhydryl ketoneCID:63950182(3). The was reacted with concentrated liquid ammonia in methanol solvent in an autoclave. The product of this step was 2-benzhydrylpyridin-3-olCID:125491889(4). Reduction of the pyridine gave 2-(diphenylmethyl)piperidin-3-olCID:209477(5). Desoxypipradrol, also known as 2-⁠diphenylmethylpiperidine (2-DPMP), is a drug developed by Ciba in the 1950s which acts as a norepinephrine-dopamine reuptake inhibitor (NDRI). Chemistry Desoxypipradrol is closely related on a structural level to the compounds methylphenidate and pipradrol, all three of which share a similar pharmacological action. Of these three piperidines, desoxypipradrol has the longest elimination half-life, as it is a highly lipophilic molecule lacking polar functional groups that are typically targeted by metabolic enzymes, giving it an extremely long duration of action when compared to most psychostimulants.
