FYVE Domains
In molecular biology the FYVE zinc finger domain is named after the four cysteine-rich proteins: Fab 1 (yeast orthologue of PIKfyve), YOTB, Vac 1 (vesicle transport protein), and EEA1, in which it has been found. FYVE domains bind phosphatidylinositol 3-phosphate, in a way dependent on its metal ion coordination and basic amino acids. The FYVE domain inserts into cell membranes in a pH-dependent manner. The FYVE domain has been connected to vacuolar protein sorting and endosome function. Structure The FYVE domain is composed of two small beta hairpins (or zinc knuckles) followed by an alpha helix. The FYVE finger binds two zinc ions. The FYVE finger has eight potential zinc coordinating cysteine positions and is characterized by having basic amino acids around the cysteines. Many members of this family also include two histidines in a sequence motif: The FYVE finger is structurally similar to the RING domain and the PHD finger. Examples The following is a list of human prote ...

ZFYVE16
Zinc finger FYVE domain-containing protein 16 is a protein that in humans is encoded by the ''ZFYVE16'' gene. The ZFYVE16 gene encodes endofin, an endosomal protein implicated in regulating membrane trafficking. It is characterized by the presence of a phosphatidylinositol 3-phosphate-binding FYVE domain positioned in the middle of the molecule (Seet et al., 2004). upplied by OMIMref name="entrez" /> In melanocytic cells ZFYVE16 gene expression may be regulated by MITF. Interactions ZFYVE16 has been shown to interact with TOM1 Target of Myb protein 1 is a protein that in humans is encoded by the ''TOM1'' gene. The specific function of this gene has not yet been determined, yet it may involve the translocation of growth factor receptor complexes to the lysosome for degra .... References Further reading

ZFYVE1
Zinc finger FYVE domain-containing protein 1 is a protein that in humans is encoded by the ''ZFYVE1'' gene. The FYVE domain mediates the recruitment of proteins involved in membrane trafficking and cell signaling to phosphatidylinositol 3-phosphate (PtdIns(3)P)-containing membranes. This gene encodes a protein which contains two zinc-binding FYVE domains in tandem. This protein displays a predominantly Golgi, endoplasmic reticulum and vesicular distribution. Alternatively spliced transcript variants have been found for this gene, and they encode two isoforms A protein isoform, or "protein variant", is a member of a set of highly similar proteins that originate from a single gene or gene family and are the result of genetic differences. RUFY2
RUN and FYVE domain containing 2 (RUFY2) is a protein that in humans is encoded by the ''RUFY2'' gene. The RUFY2 gene is named for two of its domains, the RUN domain and FYVE domains. RUFY2 is a member of the RUFY family of proteins that include RUFY1, RUFY2, RUFY3, and RUFY4. RUFY2 protein has a dynamic role in endosomal membrane trafficking. Gene The human ''RUFY2'' gene is located on the long (q) arm of chromosome 10 at region 21 band 3, from base pair 70,100,864 to base pair 70,167,051 on the reverse strand (Build GRCh37/hg19(map) The gene produces a 2,080 base pair mRNA. There are 18 predicted exons in the human gene with 13 alternative transcripts. Gene neighborhood 8,180 base pairs upstream of RUFY2 is the protein-coding gene for phenazine biosynthesis-like protein domain containing (PBLD).
