A1 Broth
An A1 broth is a liquid culture medium used in microbiology for the detection of fecal coliforms in foods, treated wastewater and seawater bays using the most probable number (MPN) method. It is prepared according to the formulation of Andrews and Presnell given below. It is used with a Durham tube, a positive tube being one that exhibits a trapped bubble of gas. Typical formula (g/L) Directions Suspend the dry ingredients in one liter of cold distilled water. Gently heat until completely dissolved and distribute 9 mL into test tubes with an inverted Durham tube. Fecal Coliforms
A fecal coliform (British: faecal coliform) is a facultatively anaerobic, rod-shaped, gram-negative, non-sporulating bacterium. Coliform bacteria generally originate in the intestines of warm-blooded animals. Most Probable Number
The most probable number method, otherwise known as the method of Poisson zeroes, is a method of getting quantitative data on concentrations of discrete items from positive/negative (incidence) data. There are many discrete entities that are easily detected but difficult to count. Any sort of amplification reaction or catalysis reaction obliterates easy quantification but allows presence to be detected very sensitively. Common examples include microorganism growth, enzyme action, or catalytic chemistry. The MPN method involves taking the original solution or sample, and subdividing it by orders of magnitude (frequently 10× or 2×), and assessing presence/absence in multiple subdivisions. The degree of dilution at which absence begins to appear indicates that the items have been diluted so much that there are many subsamples in which none appear. Durham Tube
Durham tubes are used in microbiology to detect production of gas by microorganisms. They are simply smaller test tubes inserted upside down in another test tube so they are freely movable. The culture media to be tested is then added to the larger tube and sterilized, which also eliminates the initial air gap produced when the tube is inserted upside down. Tryptone is the assortment of peptides formed by the digestion of casein by the protease trypsin. Triton X-100
Triton X-100 (''n'') is a nonionic surfactant that has a hydrophilic polyethylene oxide chain (on average it has 9.5 ethylene oxide units) and an aromatic hydrocarbon lipophilic or hydrophobic group. The hydrocarbon group is a 4-( 1,1,3,3-tetramethylbutyl)-phenyl group. Triton X-100 is closely related to IGEPAL CA-630, which might differ from it mainly in having slightly shorter ethylene oxide chains.
