Pentylone
Pentylone (β-Keto-Methylbenzodioxolylpentanamine, βk-Methyl-K, βk-MBDP, methylenedioxypentedrone, or 1‐(3,4‐methylenedioxyphenyl)‐2‐(methylamino)pentan‐1‐one) is a stimulant developed in the 1960s. It is a substituted cathinone (a type of substituted phenethylamine). It has been identified in some samples of powders sold as "NRG-1", along with varying blends of other cathinone derivatives including flephedrone, MDPBP, MDPV and 4-MePPP. It was also found in combination with 4-MePPP being sold as "NRG-3". Reports indicate side effects include feelings of paranoia, agitation and inability to sleep, with effects lasting for several days at high doses. Pharmacology Pentylone acts as a serotonin-norepinephrine-dopamine reuptake inhibitor and a serotonin releasing agent. Dipentylone
Dipentylone (also known as Dimethylpentylone) is a substituted cathinone derivative with stimulant effects, which has been sold as a designer drug, first detected in Sweden in 2014. N,N-DiMethylPentylone was first identified in toxicology samples in the USA in late 2021 and has been tracked by The Center for Forensic Science Research and Education (CFSRE) as an increasing common drug mismarketed as MDMA where Eutylone was previously commonly mismarketed as MDMA. Dimethylpentylone is not specifically listed in the United States Controlled Substance Act but would likely be considered illegal as a positional isomer of N-Ethylpentylone which is a schedule 1 substance in the United States and defined as "all isomers" within that definition. 3',4'-Methylenedioxy-α-pyrrolidinobutyrophenone (MDPBP) is a stimulant of the cathinone class developed in the 1960s, which has been reported as a novel designer drug. MDPBP is sometimes sold under the name "NRG-1" as a mixture with other cathinone derivatives, including flephedrone, pentylone, MαPPP and its higher homologue MDPV. As with other cathinones, MDPBP has been shown to have reinforcing effects in rats. The main metabolic steps are thought to be demethylenation followed by methylation of one hydroxy group, aromatic and side chain hydroxylation, oxidation of the pyrrolidine ring to the corresponding lactam as well as ring opening to the corresponding carboxylic acid. CYP2C19 and CYP2D6 have been identified as the isoenzymes mainly responsible for demethylenation. Legal Status MDPBP is a controlled substance in Poland and China. Pentedrone
Pentedrone (also known as α-methylaminovalerophenone) is a stimulant of the cathinone class that has been sold as a designer drug and has been found since 2010 as an ingredient in a number of " bath salt" mixes sold as legal highs. Pharmacology Pentedrone acts as a norepinephrine-dopamine reuptake inhibitor without causing their release, the same mechanism of action as methylphenidate. Side effects Pentedrone has been linked to at least one death where it was combined with α-PVP and caused heart failure. Detection A forensic standard of Pentedrone is available, and the compound has been posted on the Forendex website of potential drugs of abuse. Legal status On January 28, 2014, the DEA listed it, along with nine other synthetic cathinones, on the Schedule 1 with a temporary ban, effective February 27, 2014. Alpha-Pyrrolidinopentiophenone
α-Pyrrolidinopentiophenone (also known as α-pyrrolidinovalerophenone, α-PVP, O-2387, β-keto-prolintane, prolintanone, or desmethylpyrovalerone) is a synthetic stimulant of the cathinone class developed in the 1960s that has been sold as a designer drug. Colloquially, it is sometimes called flakka. α-PVP is chemically related to pyrovalerone and is the ketone analog of prolintane.
