Coluracetam (INN; development code BCI-540; formerly MKC-231) is a purported nootropic agent of the racetam family. It is contains a chemical group that is a bioisostere of the 9-amino-tetrahydroacridine family. It was initially developed and tested by the Mitsubishi Tanabe Pharma Corporation for Alzheimer's disease. Racetam
Racetams are a class of drugs that share a pyrrolidone nucleus. Some, such as piracetam, aniracetam, oxiracetam, pramiracetam and phenylpiracetam are considered nootropics. Others such as levetiracetam, brivaracetam, and seletracetam are anticonvulsants. Mechanism There is no universally accepted mechanism of action for racetams. Racetams generally show negligible affinity for common central nervous system receptors, but modulation of central neurotransmitters, including acetylcholine and glutamate, has been reported. Although aniracetam and nebracetam show affinity for muscarinic receptors, only nefiracetam demonstrates nanomolar interactions. Tricyanoaminopropene
Tricyanoaminopropene (TRIAP, TCAP, Malononitrile Dimer, 1,1,3-tricyano-2-amino-1-propene) is a nootropic drug which mimics the function of nerve growth factor and increases the growth of nerves and tissue regeneration both in isolated tissues and ''in vivo''. It stimulates the action of the enzyme choline acetyltransferase, resulting in increased acetylcholine production. This then results in increased synthesis of RNA in many different tissues in the body. However it also suppresses the production of thyroxine, causing temporary hypothyroidism which returns to normal once the drug is discontinued. Tricyanoaminopropene reduces the amnesia produced by electroconvulsive shock, and animal tests suggested nootropic activity, but no beneficial effect was found when it was tested in mentally disabled children, and administration to pregnant rats actually reduced learning ability in their young because of its anti-thyroid hormone effects. Piracetam is a drug marketed as a treatment for myoclonus. It is also used as a cognitive enhancer to improve memory, attention, and learning.
