Jonathan Marc Rothberg (born April 28, 1963) is an American scientist and entrepreneur. He is best known for his contributions to next-generation DNA sequencing. He works and resides in Guilford, Connecticut. Early life Rothberg was born in New Haven, Connecticut to Lillian Rothberg and Henry Rothberg, a chemical engineer. Prior to Rothberg's birth, his parents founded Laticrete International, Inc. a family-owned manufacturer of products for the installation of tile and stone. As a child Jonathan went on sales calls with his father. Rothberg's family laid the foundation for his scientific career. Education and scientific career Rothberg earned a BS in chemical engineering with an option in biomedical engineering from Carnegie Mellon University in 1985. Ion Semiconductor Sequencing
Ion semiconductor sequencing is a method of DNA sequencing based on the detection of hydrogen ions that are released during the polymerization of DNA. This is a method of "sequencing by synthesis", during which a complementary strand is built based on the sequence of a template strand. A microwell containing a template DNA strand to be sequenced is flooded with a single species of deoxyribonucleotide triphosphate (dNTP). If the introduced dNTP is complementary to the leading template nucleotide, it is incorporated into the growing complementary strand. This causes the release of a hydrogen ion that triggers an ISFET ion sensor, which indicates that a reaction has occurred. If homopolymer repeats are present in the template sequence, multiple dNTP molecules will be incorporated in a single cycle. Lincoln David Stein is a scientist and Professor in bioinformatics and computational biology at the Ontario Institute for Cancer Research. The GenBank sequence database is an open access, annotated collection of all publicly available nucleotide sequences and their protein translations. Massive parallel sequencing or massively parallel sequencing is any of several high-throughput approaches to DNA sequencing using the concept of massively parallel processing; it is also called next-generation sequencing (NGS) or second-generation sequencing.
