Lymphatic Endothelium
The lymphatic endothelium is a specialised form of epithelium, distinct from but similar to vascular endothelium. A lymph capillary endothelial cell is distinct from other endothelial cells in that collagen fibers are directly attached to its plasma membrane. Although lymphatics were first described by Hippocrates in 400BC and rediscovered as "milky veins in the gut of a well fed dog" in the 17th century by Gasparo Aselli, they were ignored for centuries until in 1937 Howard Florey showed that lymphatics enlarge in inflammation. At this stage vascular and lymphatic endothelia were seen to be morphologically distinct and lymphatic vessels considered less important. Later it was discovered that VEGF-R3 and VEGF-C/VEGF-D were the key growth factors controlling lymphatic endothelial proliferation. Epithelium
Epithelium or epithelial tissue is one of the four basic types of animal tissue, along with connective tissue, muscle tissue and nervous tissue. It is a thin, continuous, protective layer of compactly packed cells with a little intercellular matrix. Epithelial tissues line the outer surfaces of organs and blood vessels throughout the body, as well as the inner surfaces of cavities in many internal organs. An example is the epidermis, the outermost layer of the skin. There are three principal shapes of epithelial cell: squamous (scaly), columnar, and cuboidal. These can be arranged in a singular layer of cells as simple epithelium, either squamous, columnar, or cuboidal, or in layers of two or more cells deep as stratified (layered), or ''compound'', either squamous, columnar or cuboidal. Howard Florey
Howard Walter Florey, Baron Florey (24 September 189821 February 1968) was an Australian pharmacologist and pathologist who shared the Nobel Prize in Physiology or Medicine in 1945 with Sir Ernst Chain and Sir Alexander Fleming for his role in the development of penicillin. Although Fleming received most of the credit for the discovery of penicillin, it was Florey who carried out the first clinical trials of penicillin in 1941 at the Radcliffe Infirmary in Oxford on the first patient, a police constable from Oxford. The patient started to recover, but subsequently died because Florey was unable, at that time, to make enough penicillin. It was Florey and Chain who actually made a useful and effective drug out of penicillin, after the task had been abandoned as too difficult. Endothelium
The endothelium is a single layer of squamous endothelial cells that line the interior surface of blood vessels and lymphatic vessels. The endothelium forms an interface between circulating blood or lymph in the lumen and the rest of the vessel wall. Endothelial cells form the barrier between vessels and tissue and control the flow of substances and fluid into and out of a tissue. Endothelial cells in direct contact with blood are called vascular endothelial cells whereas those in direct contact with lymph are known as lymphatic endothelial cells. Vascular endothelial cells line the entire circulatory system, from the heart to the smallest capillaries. PDPN
Podoplanin is a protein that in humans is encoded by the ''PDPN'' gene. Structure and function Podoplanin is a mucin-type protein with a mass of 36- to 43-kDa. It is relatively well conserved between species, with homologues in humans, mice, rats, dogs and hamsters. This gene encodes a type-I, integral membrane, heavily O-glycosylated glycoprotein with diverse distribution in human tissues. The physiological function of this protein may be related to its mucin-type character. The homologous protein in other species has been described as a differentiation antigen and influenza-virus receptor. The specific function of this protein has not been determined but it has been proposed as a marker of lung injury. VEGF-C
Vascular endothelial growth factor C (VEGF-C) is a protein that is a member of the platelet-derived growth factor / vascular endothelial growth factor (PDGF/VEGF) family. It is encoded in humans by the ''VEGFC'' gene, which is located on chromosome 4q34. Functions The main function of VEGF-C is to promote the growth of lymphatic vessels (lymphangiogenesis). It acts on lymphatic endothelial cells (LECs) primarily via its receptor VEGFR-3 promoting survival, growth and migration. It was discovered in 1996 as a ligand for the orphan receptor VEGFR-3. Gasparo Aselli
Gaspare Aselli (or Asellio) ( – 9 September 1625). was an Italian physician noted for the discovery of the lacteal vessels of the lymphatic system. Aselli discovered (or rediscovered) the chylous vessels, and studied systematically the significance of these vascular structures. Life Gaspare Aselli was born in Cremona of a wealthy patrician family. He attended the University of Pavia, where he obtained degrees in medicine, surgery and philosophy. He continued his studies in Milan, where he practiced medicine with great distinction. The Milanese Senate named him an honorary citizen “by virtue of his superlative qualities as doctor and scientist”. In 1612 he was appointed Head Surgeon of the Spanish Armada in Italy. He became professor of anatomy and surgery at the University of Pavia shortly before his death in 1625 at the age of 44. Lymph (from Latin, , meaning "water") is the fluid that flows through the lymphatic system, a system composed of lymph vessels (channels) and intervening lymph nodes whose function, like the venous system, is to return fluid from the tissues to be recirculated. At the origin of the fluid-return process, interstitial fluid—the fluid between the cells in all body tissues—enters the lymph capillaries.
