Klebsiella Oxytoca
''Klebsiella oxytoca'' is a Gram-negative, rod-shaped bacterium that is closely related to ''K. pneumoniae'', from which it is distinguished by being indole-positive; it also has slightly different growth characteristics in that it is able to grow on melezitose, but not 3-hydroxybutyrate. It was first described in 1886 when it was isolated from sour milk and named ''Bacillus oxytocus perniciosus'' (from Greek ''oxus'' 'sour' + ''-tokos'' 'producing'). ''Klebsiella oxytoca'' is characterized by negative methyl red, positive VP, positive citrate, urea and TSI gas production, is AA, and negative for TSI sulfide, DNAse, growth on sulfide-indole motility medium and the phenylalanine deaminase test. It is a diazotroph, able to colonise plant hosts and fix atmospheric nitrogen into a form which the plant can use. Carl Flügge
Carl Georg Friedrich Wilhelm Flügge (12 September 1847 – 10 December 1923) was a German bacteriologist and hygienist. His finding that pathogens were present in expiratory droplets, the eponymous Flügge droplets, laid ground for the concept of droplet transmission as a route for the spread of respiratory infectious diseases. Early life and education Carl Flügge was a native of Hanover. He studied medicine in Göttingen, Bonn, Leipzig and Munich, and in 1878 was a lecturer of hygiene in Berlin. Career In 1881 Flügge became the first chair of hygiene at the University of Göttingen, and afterwards a professor at the Universities of Breslau and Berlin, where he succeeded Max Rubner at the Department of Hygiene. Diazotrophs are bacteria and archaea that fix gaseous nitrogen in the atmosphere into a more usable form such as ammonia. Citrate Test
The citrate test detects the ability of an organism to use citrate as the sole source of carbon and energy. Principle Bacteria are inoculated on a medium containing sodium citrate and a pH indicator such as bromothymol blue. The medium also contains inorganic ammonium salts, which are utilized as sole source of nitrogen. Use of citrate involves the enzyme citrate lyase, which breaks down citrate to oxaloacetate and acetate. Oxaloacetate is further broken down to pyruvate and carbon dioxide (CO2). Production of sodium bicarbonate (NaHCO3) as well as ammonia (NH3) from the use of sodium citrate and ammonium salts results in alkaline pH. This results in a change of the medium's color from green (neutral) to blue (alkaline). Voges–Proskauer Test
Voges–Proskauer or VP is a test used to detect acetoin in a bacterial broth culture. The test is performed by adding 1-Naphthol, alpha-naphthol and potassium hydroxide to the Voges-Proskauer broth, which is a Glucose phosphate broth, glucose-phosphate broth that has been inoculated with bacteria. A cherry red color indicates a positive result, while a yellow-brown color indicates a negative result. The test depends on the digestion of glucose to acetylmethylcarbinol. In the presence of oxygen and strong base, the acetylmethylcarbinol is oxidized to diacetyl, which then reacts with guanidine compounds commonly found in the peptone medium of the broth. Alpha-naphthol acts as a color enhancer, but the color change to red can occur without it. Procedure: First, add the alpha-naphthol; then, add the potassium hydroxide.
