Chromosome Conformation Capture
Chromosome conformation capture techniques (often abbreviated to 3C technologies or 3C-based methods) are a set of molecular biology methods used to analyze the spatial organization of chromatin in a cell. These methods quantify the number of interactions between genomic loci that are nearby in 3-D space, but may be separated by many nucleotides in the linear genome. Such interactions may result from biological functions, such as promoter- enhancer interactions, or from random polymer looping, where undirected physical motion of chromatin causes loci to collide. Interaction frequencies may be analyzed directly, or they may be converted to distances and used to reconstruct 3-D structures. The chief difference between 3C-based methods is their scope. For example, when using PCR to detect interaction in a 3C experiment, the interactions between two specific fragments are quantified. A chromosome is a long DNA molecule with part or all of the genetic material of an organism. In most chromosomes the very long thin DNA fibers are coated with packaging proteins; in eukaryotic cells the most important of these proteins are the histones. These proteins, aided by chaperone proteins, bind to and condense the DNA molecule to maintain its integrity. These chromosomes display a complex three-dimensional structure, which plays a significant role in transcriptional regulation. Chromosomes are normally visible under a light microscope only during the metaphase of cell division (where all chromosomes are aligned in the center of the cell in their condensed form). Before this happens, each chromosome is duplicated (S phase), and both copies are joined by a centromere, resulting either in an X-shaped structure (pictured above), if the centromere is located equatorially, or a two-arm structure, if the centromere is located distally. Emil Heitz
Emil Heitz (born 19 October 1892 in Strasbourg, then part of the German Empire - died 8 July 1965 in Lugano, Switzerland) was a German-Swiss botanist and geneticist. In 1928, Heitz detected differences along chromosomes that correlated with linear arrangement of genes, suggesting the terms euchromatin and heterochromatin for differences detectable by suitable chromosomal stains. His subsequent studies developed what he called cytological genetics, laying the ground for what is now termed cytogenetics. In November 1933 Heitz was a signatory of the ''Vow of allegiance of the Professors of the German Universities and High-Schools to Adolf Hitler and the National Socialistic State''. In cell biology, chromosome territories are regions of the nucleus preferentially occupied by particular chromosomes. Interphase chromosomes are long DNA strands that are extensively folded, and are often described as appearing like a bowl of spaghetti. The chromosome territory concept holds that despite this apparent disorder, chromosomes largely occupy defined regions of the nucleus. Nucleosome
A nucleosome is the basic structural unit of DNA packaging in eukaryotes. The structure of a nucleosome consists of a segment of DNA wound around eight histone proteins and resembles thread wrapped around a spool. The nucleosome is the fundamental subunit of chromatin. Each nucleosome is composed of a little less than two turns of DNA wrapped around a set of eight proteins called histones, which are known as a histone octamer. Each histone octamer is composed of two copies each of the histone proteins H2A, H2B, H3, and H4. DNA must be compacted into nucleosomes to fit within the cell nucleus. Pierre Chambon (born 7 February 1931 in Mulhouse, France) was the founder of the in Strasbourg, France. X-inactivation (also called Lyonization, after English geneticist Mary Lyon) is a process by which one of the copies of the X chromosome is inactivated in therian female mammals. The inactive X chromosome is silenced by being packaged into a transcriptionally inactive structure called heterochromatin.
