Operational Taxonomic Unit
An operational taxonomic unit (OTU) is an operational definition used to classify groups of closely related individuals. The term was originally introduced in 1963 by Robert R. Sokal and Peter H. A. Sneath in the context of numerical taxonomy, where an "operational taxonomic unit" is simply the group of organisms currently being studied. In this sense, an OTU is a pragmatic definition to group individuals by similarity, equivalent to but not necessarily in line with classical Linnaean taxonomy or modern evolutionary taxonomy. Nowadays, however, the term "OTU" is commonly used in a different context and refers to clusters of (uncultivated or unknown) organisms, grouped by DNA sequence similarity of a specific taxonomic marker gene (originally coined as mOTU; molecular OTU). An operational definition specifies concrete, replicable procedures designed to represent a construct. In the words of American psychologist S.S. Stevens (1935), "An operation is the performance which we execute in order to make known a concept." For example, an operational definition of "fear" (the construct) often includes measurable physiologic responses that occur in response to a perceived threat. Thus, "fear" might be operationally defined as specified changes in heart rate, electrodermal activity, pupil dilation, and blood pressure. 18S ribosomal RNA (abbreviated 18S rRNA) is a part of the ribosomal RNA in eukaryotes. It is a component of the Eukaryotic small ribosomal subunit (40S) and the cytosolic homologue of both the 12S rRNA in mitochondria and the 16S rRNA in plastids and prokaryotes. Similar to the prokaryotic 16S rRNA, the genes of the 18S ribosomal RNA have been widely used for phylogenetic studies and biodiversity screening of eukaryotes. Phylotype

An amplicon sequence variant (ASV) is any one of the inferred single DNA sequences recovered from a high-throughput analysis of marker genes. Because these analyses, also called "amplicon reads," are created following the removal of erroneous sequences generated during PCR and sequencing, using ASVs makes it possible to distinguish sequence variation by a single nucleotide change. The uses of ASVs include classifying groups of species based on DNA sequences, finding biological and environmental variation, and determining ecological patterns. 16S RRNA
16S ribosomal RNA (or 16Svedberg, S rRNA) is the RNA component of the 30S subunit of a prokaryotic ribosome (SSU rRNA). It binds to the Shine-Dalgarno sequence and provides most of the SSU structure. The genes coding for it are referred to as 16S rRNA genes and are used in reconstructing phylogenetics, phylogenies, due to the slow rates of evolution of this region of the gene. Carl Woese and George E. Fox were two of the people who pioneered the use of 16S rRNA in phylogenetics in 1977. Multiple sequences of the 16S rRNA gene can exist within a single bacterium. Robert Lee Rayford (February 3, 1953 – May 15, 1969), sometimes identified as Robert R. due to his age, was an American teenager from Missouri who has been suggested to represent the earliest confirmed case of HIV/AIDS in North America. This is based on evidence published in 1988 in which the authors claimed that medical evidence indicated that he was "infected with a virus closely related or identical to human immunodeficiency virus type 1." Rayford died of pneumonia, but his other symptoms baffled the doctors who treated him. A study published in 1988 reported the detection of antibodies against HIV. Results of testing for HIV genetic material were reported at a scientific conference in Australia in 1999. However, the data has never been published in a peer-reviewed medical or scientific journal.
