''Clostridioides'' is a genus of Gram-positive bacteria, which includes ''Clostridioides difficile'', a human pathogen causing an infectious diarrhea. Taxonomy The genus ''Clostridioides'' was created to describe a few species formerly in the genus ''Clostridium'' which have been shown to be their own genetically distinct genus using 16S rRNA gene sequencing analysis. However, both names are still in use and valid under the International Code of Nomenclature of Prokaryotes. Since '' C. mangenotii'' was further separated into a distinct genus in 2024, ''Clostridioides'' is a monotypic genus. Description They are obligate anaerobes capable of producing endospores. ''Clostridioides difficile'' ( syn. ''Clostridium difficile'') is a bacterium known for causing serious diarrheal infections, and may also cause colon cancer. It is known also as ''C. difficile'', or ''C. diff'' (), and is a Gram-positive species of spore-forming bacteria. ''Clostridioides'' spp. are anaerobic, motile bacteria, ubiquitous in nature and especially prevalent in soil. Its vegetative cells are rod-shaped, pleomorphic, and occur in pairs or short chains. Under the microscope, they appear as long, irregular (often drumstick- or spindle-shaped) cells with a bulge at their terminal ends (forms subterminal spores). Under Gram staining, ''C. difficile'' cells are Gram-positive and show optimum growth on blood agar at human body temperatures in the absence of oxygen. The Peptostreptococcaceae are a family of Gram-positive anaerobic bacteria in the class Clostridia. A majority of members are identified as obligate anaerobes. In bacteriology, gram-positive bacteria are bacteria that give a positive result in the Gram stain test, which is traditionally used to quickly classify bacteria into two broad categories according to their type of cell wall. The Gram stain is used by microbiologists to place bacteria into two main categories, gram-positive (+) and gram-negative (−). Gram-positive bacteria have a thick layer of peptidoglycan within the cell wall, and gram-negative bacteria have a thin layer of peptidoglycan. Gram-positive bacteria retain the crystal violet stain used in the test, resulting in a purple color when observed through an optical microscope. The thick layer of peptidoglycan in the bacterial cell wall retains the stain after it has been fixed in place by iodine.
