Anterograde Tracing
In neuroscience, anterograde tracing is a research method that is used to trace axonal projections from their source (the cell body, or soma) to their point of termination (the synapse). A hallmark of anterograde tracing is the labeling of the presynaptic and the postsynaptic neuron(s). The crossing of the synaptic cleft is a vital difference between the anterograde tracers and the dye fillers used for morphological reconstruction. The complementary technique is ''retrograde tracing'', which is used to trace neural connections from their termination to their source (i.e. synapse to cell body). Both the anterograde and retrograde tracing techniques are based on the visualization of the biological process of axonal transport. Viral neuronal tracing is the use of a virus to trace neural pathways, providing a self-replicating tracer. Viruses have the advantage of self-replication over molecular tracers but can also spread too quickly and cause degradation of neural tissue. Viruses that can infect the nervous system, called neurotropic viruses, spread through spatially close assemblies of neurons through synapses, allowing for their use in studying functionally connected neural networks. Phytohaemagglutinin
Phytohaemagglutinin (PHA, or phytohemagglutinin) is a lectin found in plants, especially certain legumes. PHA actually consists of two closely related proteins, called leucoagglutinin (PHA-L) and PHA-E. These proteins cause blood cells to clump together. PHA-E cause erythrocytes (red blood cells) to clump. PHA-L causes leukocytes (white blood cells) to clump. Phytohaemagglutinin has carbohydrate-binding specificity for a complex oligosaccharide containing galactose, ''N''-acetylglucosamine, and mannose. It is found in the highest concentrations in uncooked red kidney beans and white kidney beans (also known as cannellini), and it is also found in lower quantities in many other types of green beans and other common beans (''Phaseolus vulgaris''), as well as broad beans (''Vicia faba'') such as fava beans. Wheat germ agglutinin (WGA) is a lectin that protects wheat (''Triticum'') from insects, yeast and bacteria. An agglutinin protein, it binds to N-acetyl-D-glucosamine and sialic acid. WGA has also been shown to interact with sialic acid residues on oligosaccharides. Succinic acid, Succinylated WGA is selective for β-N-acetylglucosamine (β-GlcNAc), making it a useful tool for detecting O-GlcNAc, ''O''-GlcNAc. WGA is composed of a mixture of three isoforms (WGA1, WGA2, WGA3), which are quite similar to each other and each contain an unusually high amount of glycine. The cytoskeleton is a complex, dynamic network of interlinking protein filaments present in the cytoplasm of all cells, including those of bacteria and archaea. In eukaryotes, it extends from the cell nucleus to the cell membrane and is composed of similar proteins in the various organisms. Nectin
Nectins and Nectin-like molecules (Necl) are families of cellular adhesion molecules involved in Calcium in biology, Ca2+-independent cellular adhesion.
