Historadiography
Historadiography is a technique formerly utilized in the fields of histology and cellular biology to provide semiquantitative information regarding the density of a tissue sample. It is usually synonymous with microradiography. This is achieved by layering a ground section of mineralized tissue (such as bone) with photographic emulsion on a glass slide and exposing the sample to a beam of X-rays. After developing the emulsion, the resulting radiograph can be viewed with a microscope. A side-by-side comparison with a slide containing radiographs of various substances of known mass can provide a rough mass estimate, and therefore a rough approximation of the concentration of calcium salts in the sample. Historadiography has also been used to visualize staining of tissue, such as spinal cord samples with thorotrast, which contains thorium that is opaque to X-rays. Thorotrast
Thorotrast is a suspension containing particles of the radioactive compound thorium dioxide, ThO2; it was used as a radiocontrast agent in clinical radiography in the 1930s to 1950s. It is no longer used clinically. Thorium compounds produce excellent images because of thorium's high opacity to X-rays (it has a high cross section for absorption). However, thorium is retained in the body, and it is radioactive, emitting harmful alpha radiation as it decays. Because the suspension offered high image quality and had virtually no immediate side-effects compared to the alternatives available at the time, Thorotrast became widely used after its introduction in 1931. Autoradiography
An autoradiograph is an image on an X-ray film or nuclear emulsion produced by the pattern of decay emissions (e.g., beta particles or gamma rays) from a distribution of a radioactive substance. Alternatively, the autoradiograph is also available as a digital image (digital autoradiography), due to the recent development of scintillation gas detectors or rare earth phosphorimaging systems. The film or emulsion is apposed to the labeled tissue section to obtain the autoradiograph (also called an autoradiogram). The ''auto-'' prefix indicates that the radioactive substance is within the sample, as distinguished from the case of historadiography or microradiography, in which the sample is marked using an external source.
