Trichrome
Trichrome staining is a histological staining method that uses two or more acid dyes in conjunction with a polyacid. Staining differentiates tissues by tinting them in contrasting colours. It increases the contrast of microscopic features in cells and tissues, which makes them easier to see when viewed through a microscope. The word '' trichrome'' means "three colours". The first staining protocol that was described as "trichrome" was Mallory's trichrome stain, which differentially stained erythrocytes to a red colour, muscle tissue to a red colour, and collagen to a blue colour. Some other trichrome staining protocols are the Masson's trichrome stain, Lillie's trichrome, and the Gömöri trichrome stain. Masson's Trichrome Stain
Masson's trichrome is a three-colour staining procedure used in histology. The recipes evolved from Claude L. Pierre Masson's (1880–1959) original formulation have different specific applications, but all are suited for distinguishing cells from surrounding connective tissue. Most recipes produce red keratin and muscle fibers, blue or green collagen and bone, light red or pink cytoplasm, and dark brown to black cell nuclei. The trichrome is applied by immersion of the fixated sample into Weigert's iron hematoxylin, and then three different solutions, labeled A, B, and C: * Weigert's hematoxylin is a sequence of three solutions: ferric chloride in diluted hydrochloric acid, hematoxylin in 95% ethanol, and potassium ferricyanide solution alkalized by sodium borate. Lillie's Trichrome
Lillie's trichrome is a combination of dyes used in histology. It is similar to Masson's trichrome stain, but it uses Biebrich scarlet for the plasma stain. It was initially published by Ralph D. Lillie in 1940. It is applied by submerging the fixated sample into the following three solutions: Weigert's iron hematoxylin working solution, Biebrich scarlet solution, and Fast Green FCF solution. The resulting stains are black cell nuclei, brown cytoplasm, red muscle and myelinated fibers, blue collagen, and scarlet erythrocytes. Phosphomolybdic acid is the heteropolymetalate with the formula . It is a yellow solid, although even slightly impure samples have a greenish coloration. It is also known as dodeca molybdophosphoric acid or PMA, is a yellow-green chemical compound that is freely soluble in water and polar organic solvents such as ethanol. Mallory's trichrome stain also called Mallory's Triple Stain is a stain utilized in histology to aid in revealing different macromolecules that make up the cell. It uses the three stains: aniline blue, acid fuchsin, and orange G. As a result, this staining technique can reveal collagen, ordinary cytoplasm, and red blood cells. It is used in examining the collagen of connective tissue. For tissues that are not directly acidic or basic, it can be difficult to use only one stain to reveal the necessary structures of interest. A combination of the three different stains in precise amounts applied in the correct order reveals the details selectively. Biebrich scarlet (C.I. 26905) is a molecule used in Lillie's trichrome. The dye was created in 1878 by the German chemist Rudolf Nietzki. Biebrich scarlet dyes are used to color hydrophobic materials like fats and oils. The dye is an illegal dye for food additives because of its carcinogenic properties. Biebrich scarlet can have harmful effects on living and non-living organisms in natural water, therefore the pollutant must be removed. Removal of the pollutant involves absorption, membrane filtration, precipitation, ozonation, fungal detachment, and electrochemical separation. Hydrogel absorbents have active sites to which the dye is held using electrostatic interactions. Acid Fuchsin
Acid fuchsin or fuchsine acid, (also called Acid Violet 19 and C.I. 42685) is an acidic magenta dye with the chemical formula C20H17N3Na2O9S3. It is a sodium sulfonate derivative of fuchsine. Acid fuchsin has wide use in histology, and is one of the dyes used in Masson's trichrome stain. This method is commonly used to stain cytoplasm and nuclei of tissue sections in the histology laboratory in order to distinguish muscle from collagen.
