Rolf Hassler
Rolf Hassler (1914–1984) was a German pathologist who made important discoveries on the pathophisiology and treatment of Parkinson's disease (PD). In 1938 he published the autopsies of PD patients that showed while the striatum and globus pallidus were mostly unaffected and the main affected structure was the substantia nigra pars compacta; it lost many neurons and also held abundant Lewy bodies. Such findings confirmed Konstantin Tretiakoff Konstantin Nikolaevitch Tretiakoff (; December 26, 1892 – 1958) was a Russian neuropathologist. Parkinson's Disease
Parkinson's disease (PD), or simply Parkinson's, is a long-term degenerative disorder of the central nervous system that mainly affects the motor system. The symptoms usually emerge slowly, and as the disease worsens, non-motor symptoms become more common. The most obvious early symptoms are tremor, rigidity, slowness of movement, and difficulty with walking. Cognitive and behavioral problems may also occur with depression, anxiety, and apathy occurring in many people with PD. Parkinson's disease dementia becomes common in the advanced stages of the disease. Those with Parkinson's can also have problems with their sleep and sensory systems. The motor symptoms of the disease result from the death of cells in the substantia nigra, a region of the midbrain, leading to a dopamine deficit. Striatum
The striatum, or corpus striatum (also called the striate nucleus), is a nucleus (a cluster of neurons) in the subcortical basal ganglia of the forebrain. The striatum is a critical component of the motor and reward systems; receives glutamatergic and dopaminergic inputs from different sources; and serves as the primary input to the rest of the basal ganglia. Functionally, the striatum coordinates multiple aspects of cognition, including both motor and action planning, decision-making, motivation, reinforcement, and reward perception. Globus Pallidus
The globus pallidus (GP), also known as paleostriatum or dorsal pallidum, is a subcortical structure of the brain. It consists of two adjacent segments, one external, known in rodents simply as the globus pallidus, and one internal, known in rodents as the entopeduncular nucleus. It is part of the telencephalon, but retains close functional ties with the subthalamus in the diencephalon – both of which are part of the extrapyramidal motor system. The globus pallidus is a major component of the basal ganglia, with principal inputs from the striatum, and principal direct outputs to the thalamus and the substantia nigra. Substantia Nigra Pars Compacta
The pars compacta (SNpc) is a portion of the ''substantia nigra'', located in the midbrain. It is formed by dopaminergic neurons and located medial to the pars reticulata. Parkinson's disease is characterized by the death of dopaminergic neurons in this region. Anatomy In humans, the nerve cell bodies of the ''pars compacta'' are coloured black by the pigment neuromelanin. The degree of pigmentation increases with age. This pigmentation is visible as a distinctive black stripe in brain sections and is the origin of the name given to this volume of the brain. Lewy Bodies
Lewy bodies are the inclusion bodies – abnormal aggregations of protein – that develop inside nerve cells affected by Parkinson's disease (PD), the Lewy body dementias ( Parkinson's disease dementia and dementia with Lewy bodies (DLB)), and some other disorders. They are also seen in cases of multiple system atrophy, particularly the parkinsonian variant (MSA-P). Lewy bodies appear as spherical masses in the cytoplasm that displace other cell components. Konstantin Tretiakoff
Konstantin Nikolaevitch Tretiakoff (; December 26, 1892 – 1958) was a Russian neuropathologist. He was born in Fergana, Uzbekistan, as a son of military physician, who was member of Pierre Bonvalot's first Pamir expedition. He studied medicine in L'Assistance Publique des Hopitaux de Paris. He received his doctorate in 1919. In his thesis he described degeneration of the ''substantia nigra'' associated with paralysis agitans (Parkinson disease). Tretiakof was first to link this anatomic structure with parkinsonism. Between 1922 and 1926 Tretiakoff worked at the Hospício de Juquery, near the city of São Paulo, Brazil. In 1931 he was appointed Chairman at the new Department of Neuropathology at the Medical Institute in Saratov, USSR, where he spent the rest of his life.
