Flossie Wong-Staal
Flossie Wong-Staal (née Wong Yee Ching, ; August 27, 1946July 8, 2020) was a Chinese-American virologist and molecular biologist. She was the first scientist to clone HIV and determine the function of its genes, which was a major step in proving that HIV is the cause of AIDS. From 1990 to 2002, she held the Florence Riford Chair in AIDS Research at the University of California, San Diego (UCSD). She was co-founder and, after retiring from UCSD, she became the chief scientific officer of Immusol, which was renamed Pharmaceuticals in 2007 when it transitioned to a drug development company focused on hepatitis C and continued as chief scientific officer. Radioimmunoprecipitation Assay Buffer
Radioimmunoprecipitation assay buffer (RIPA buffer) is a lysis buffer used to lyse cells and tissue for the radio immunoprecipitation assay (RIPA). This buffer is more denaturing than NP-40 or Triton X-100 because it contains the ionic detergents SDS and sodium deoxycholate as active constituents and is particularly useful for disruption of nuclear membranes in the preparation of nuclear extracts. The stronger detergents in RIPA buffer (such as SDS) cause greater protein denaturation and decrease protein-protein interactions. The endothelium is a single layer of squamous endothelial cells that line the interior surface of blood vessels and lymphatic vessels. The endothelium forms an interface between circulating blood or lymph in the lumen and the rest of the vessel wall. Endothelial cells form the barrier between vessels and tissue and control the flow of substances and fluid into and out of a tissue. Endothelial cells in direct contact with blood are called vascular endothelial cells whereas those in direct contact with lymph are known as lymphatic endothelial cells. Kaposi's sarcoma (KS) is a type of cancer that can form masses in the skin, in lymph nodes, in the mouth, or in other organs. The skin lesions are usually painless, purple and may be flat or raised. Tat (HIV)
In molecular biology, Tat is a protein that is encoded for by the ''tat'' gene in HIV-1. Tat is a regulatory protein that drastically enhances the efficiency of viral transcription. Tat stands for "Trans-Activator of Transcription". The protein consists of between 86 and 101 amino acids depending on the subtype. Tat vastly increases the level of transcription of the HIV dsDNA. Before Tat is present, a small number of RNA transcripts will be made, which allow the Tat protein to be produced. Tat then binds to cellular factors and mediates their phosphorylation, resulting in increased transcription of all HIV genes, providing a positive feedback cycle. Genetic linkage is the tendency of DNA sequences that are close together on a chromosome to be inherited together during the meiosis phase of sexual reproduction. Retroviruses
A retrovirus is a type of virus that inserts a DNA copy of its RNA genome into the DNA of a host cell that it invades, thus changing the genome of that cell.
