Griffith's Experiment
Griffith's experiment, reported in 1928 by Frederick Griffith, was the first experiment suggesting that bacteria are capable of transferring genetic information through a process known as transformation. Griffith's findings were followed by research in the late 1930s and early 40s that isolated DNA as the material that communicated this genetic information. Pneumonia was a serious cause of death in the wake of the post-WWI Spanish influenza pandemic, and Griffith was studying the possibility of creating a vaccine. Griffith used two strains of pneumococcus ('' Diplococcus pneumoniae'') bacteria which infect mice – a type III-S (smooth) which was virulent, and a type II-R (rough) strain which was nonvirulent. The III-S strain synthesized a polysaccharide capsule that protected itself from the host's immune system, resulting in the death of the host, while the II-R strain did not have that protective capsule and was defeated by the host's immune system. Virulent
Virulence is a pathogen's or microorganism's ability to cause damage to a host. In most, especially in animal systems, virulence refers to the degree of damage caused by a microbe to its host. The pathogenicity of an organism—its ability to cause disease—is determined by its virulence factors. In the specific context of gene for gene systems, often in plants, virulence refers to a pathogen's ability to infect a resistant host. The noun ''virulence'' derives from the adjective ''virulent'', meaning disease severity. The word ''virulent'' derives from the Latin word ''virulentus'', meaning "a poisoned wound" or "full of poison." The history of genetics dates from the classical era with contributions by Pythagoras, Hippocrates, Aristotle, Epicurus, and others. Hershey–Chase Experiment
The Hershey–Chase experiments were a series of experiments conducted in 1952 by Alfred Hershey and Martha Chase that helped to confirm that DNA is genetic material. While DNA had been known to biologists since 1869, many scientists still assumed at the time that proteins carried the information for inheritance because DNA appeared to be an inert molecule, and, since it is located in the nucleus, its role was considered to be phosphorus storage. In their experiments, Hershey and Chase showed that when bacteriophages, which are composed of DNA and protein, infect bacteria, their DNA enters the host bacterial cell, but most of their protein does not. Hershey and Chase and subsequent discoveries all served to prove that DNA is the hereditary material. Quellung Reaction
The quellung reaction, also called the Neufeld reaction, is a biochemical reaction in which antibodies bind to the bacterial capsule of ''Streptococcus pneumoniae'', ''Klebsiella pneumoniae'', ''Neisseria meningitidis'', '' Bacillus anthracis'', ''Haemophilus influenzae'', ''Escherichia coli'', and '' Salmonella''. The antibody reaction allows these species to be visualized under a microscope. If the reaction is positive, the capsule becomes opaque and appears to enlarge. Fred Neufeld
Fred (Friedrich) Neufeld (17 February 1869 – 18 April 1945) was a physician and bacteriologist who discovered the pneumococcal types. This discovery led Fred Griffith to show that one pneumococcal type could be transformed into another (Griffith's experiment). Subsequently, Oswald Avery demonstrated that the transforming substance was DNA. All modern molecular biology has evolved from this work. Early years Neufeld was the son of a physician.
