Twelve O'clock Knob is located adjacent to Poor Mountain in Roanoke County directly south of Salem, Virginia. The north slope of the ridgeline formed by Poor Mountain and Twelve O'clock Knob marks the southwestern boundary of the Roanoke Valley.
