Yeast Estrogen Assay
Yeast estrogen screen (YES) and Yeast androgen screen (YAS) are ''in vitro'' screens that have been developed in order to detect estrogenic and androgenic activities, respectively, of natural and synthetic compounds, mixtures and environmental samples. The test is based on genetically modified yeast cells (''Saccharomyces cerevisiae''), which contain the gene for the human estrogen or androgen receptor coupled to a reporter gene (e.g. lacZ). Activation of the receptor initiates a cascade of events leading to the expression of the reporter gene product (e.g. β-galctosidase) which converts a substrate (e.g. CPRG) which leads to a measurable color change from yellow to red in the medium.Routledge, E.J. and Sumpter, J.P. Estrogen
Estrogen or oestrogen is a category of sex hormone responsible for the development and regulation of the female reproductive system and secondary sex characteristics. There are three major endogenous estrogens that have estrogenic hormonal activity: estrone (E1), estradiol (E2), and estriol (E3). Estradiol, an estrane, is the most potent and prevalent. Another estrogen called estetrol (E4) is produced only during pregnancy. Estrogens are synthesized in all vertebrates and some insects. An androgen (from Greek ''andr-'', the stem of the word meaning "man") is any natural or synthetic steroid hormone that regulates the development and maintenance of male characteristics in vertebrates by binding to androgen receptors. This includes the embryological development of the primary male sex organs, and the development of male secondary sex characteristics at puberty.
