Toxic Vacuolation
Toxic vacuolation, also known as toxic vacuolization, is the formation of Vacuole#Animals, vacuoles in the cytoplasm of neutrophils in response to severe infections or inflammation, inflammatory conditions. Clinical significance Toxic vacuolation is associated with sepsis, particularly when accompanied by toxic granulation. The finding is also associated with bacterial infection, alcohol toxicity, liver failure, and treatment with G-CSF#Medical use, granulocyte colony-stimulating factor, a cytokine drug used to increase the absolute neutrophil count in patients with neutropenia. The formation of toxic vacuoles represents increased phagocytosis, phagocytic activity, which is stimulated by the release of cytokines in response to inflammation or tissue injury. Toxic vacuolation frequently occurs in conjunction with toxic granulation and Döhle bodies in inflammatory states, and these findings are collectively referred to as ''toxic changes''. Neutrophils (also known as neutrocytes or heterophils) are the most abundant type of granulocytes and make up 40% to 70% of all white blood cells in humans. They form an essential part of the innate immune system, with their functions varying in different animals. Acute-phase proteins (APPs) are a class of proteins whose concentrations in blood plasma either increase (positive acute-phase proteins) or decrease (negative acute-phase proteins) in response to inflammation. Leukemoid Reaction
The term leukemoid reaction describes an increased white blood cell count (> 50,000 cells/μL), which is a physiological response to stress or infection (as opposed to a primary blood malignancy, such as leukemia). It often describes the presence of immature cells such as myeloblasts or red blood cells with nuclei in the peripheral blood. It may be lymphoid or myeloid. Jordans' Anomaly
Jordans' anomaly (also known as Jordan anomaly and Jordans bodies) is a familial abnormality of white blood cell morphology. Individuals with this condition exhibit persistent vacuolation of granulocytes and monocytes in the peripheral blood and bone marrow. Jordans' anomaly is associated with neutral lipid storage diseases. Genetics Jordans' anomaly is a characteristic finding in Chanarin-Dorfman syndrome and other neutral lipid storage diseases. The anomaly is associated with mutations in the ''PNPLA2'' gene, which produces the enzyme adipose triglyceride lipase (ATGL), and the ''ABHD5'' gene, which encodes a cofactor of ATGL. Neutral lipid storage disease (also known as Chanarin–Dorfman syndrome) is a congenital autosomal recessive disorder characterized by accumulation of triglycerides in the cytoplasm of leukocytes[1], Jordans anomaly, (Jordan’s Anomaly) muscle, liver, fibroblasts, and other tissues. Band Neutrophil
A band cell (also called band neutrophil, band form or stab cell) is a cell undergoing granulopoiesis, derived from a metamyelocyte, and leading to a mature granulocyte. It is characterized by having a curved but not lobular nucleus. The term "band cell" implies a granulocytic lineage (e.g., neutrophils). Clinical significance Band neutrophils are an intermediary step prior to the complete maturation of segmented neutrophils. Polymorphonuclear neutrophils are initially released from the bone marrow as band cells, as the immature neutrophils become activated or exposed to pathogens, their nucleus will take on a segmented appearance. Left shift or blood shift is an increase in the number of immature cell types among the blood cells in a sample of blood.
