Intracerebroventricular Injection
Intracerebroventricular injection (also called ICV injection, i.c.v. injection, or sometimes ICVI) is an invasive injection technique of substances directly into the cerebrospinal fluid in cerebral ventricles in order to bypass the bloodbrain barrier. Although this barrier effectively protects the brain, it can prevent important medications from entering the CNS. The technique is widely used in biomedical research to introduce drugs, therapeutic RNAs, plasmid DNAs, and viral vectors into the CNS of diseased mice models. It can also be used in human in cases of neurodegenerative disorders like spinal muscular atrophy (SMA), or administering chemotherapy in gliomas as well as delivering neurotrophic factors to CNS. It can be contrasted with intraperitoneal injection as an alternative choice of route of administration with differing pharmacokinetic and pharmacodynamic effects. Cerebrospinal Fluid
Cerebrospinal fluid (CSF) is a clear, colorless body fluid found within the tissue that surrounds the brain and spinal cord of all vertebrates. CSF is produced by specialised ependymal cells in the choroid plexus of the ventricles of the brain, and absorbed in the arachnoid granulations. There is about 125 mL of CSF at any one time, and about 500 mL is generated every day. CSF acts as a shock absorber, cushion or buffer, providing basic mechanical and immunological protection to the brain inside the skull. CSF also serves a vital function in the cerebral autoregulation of cerebral blood flow. CSF occupies the subarachnoid space (between the arachnoid mater and the pia mater) and the ventricular system around and inside the brain and spinal cord. The lateral ventricles are the two largest ventricular system, ventricles of the brain and contain cerebrospinal fluid (CSF). Each cerebral hemisphere contains a lateral ventricle, known as the left or right ventricle, respectively. Ayub Ommaya
Ayub Khan Ommaya MD, ScD (hc), FRCS, FACS (1930 - 2008) was a Pakistani American neurosurgeon and the inventor of the Ommaya reservoir. The reservoir is used to provide chemotherapy directly to the tumor site for brain tumors. Ommaya was also a leading expert in traumatic brain injuries. Ommaya published over 150 articles, chapters, and books. His research focused on cancer treatment, traumatic brain injury, a CSF artificial organ, and philosophy of mind. Through discussions with Congressman William Lehman Chair of the House Appropriations Committee responsible for the Department of Transportation, he developed CDC's National Center for Injury Prevention and Control, which as part of its mission focuses on traumatic brain injury. Ommaya Reservoir
An Ommaya reservoir is an intraventricular catheter system that can be used for the aspiration of cerebrospinal fluid or for the delivery of drugs (e.g. chemotherapy) into the cerebrospinal fluid. It consists of a catheter in one lateral ventricle attached to a reservoir implanted under the scalp. It is used to treat brain tumors, leukemia/lymphoma or leptomeningeal disease by intrathecal drug administration. Intraperitoneal injection or IP injection is the injection of a substance into the peritoneum (body cavity). It is more often applied to animals than to humans. In general, it is preferred when large amounts of blood replacement fluids are needed or when low blood pressure or other problems prevent the use of a suitable blood vessel for intravenous injection. The ventricular system is a set of four interconnected cavities known as cerebral ventricles in the brain. Within each ventricle is a region of choroid plexus which produces the circulating cerebrospinal fluid (CSF).
