Pharmacyclics LLC is an American biopharmaceutical company based in Sunnyvale, California. It is primarily focused on the development of cancer therapies. In 2017, Xynomic Pharmaceuticals acquired all global rights to Abexinostat, Pharmacyclics' primary commercial product. Acquisition by AbbVie In March 2015, Chicago-based biopharmaceutical firm AbbVie announced it would acquire Pharmacyclics, as well as its lead anti-cancer compound ibrutinib (Imbruvica) for $21 billion. As part of the deal, AbbVie will pay $261.25 per share in a mixture of both cash and AbbVie equity. The merger is expected to close in mid-2015. Robert Duggan (venture Capitalist)
Robert W. Duggan (born 1944) is an American billionaire, entrepreneur, philanthropist, biotech executive, health care executive, educator, and human rights activist. He is the former CEO of biopharmaceutical company Pharmacyclics and was previously CEO of surgical systems maker Computer Motion from 1997 to 2003. Ibrutinib
Ibrutinib, sold under the brand name Imbruvica among others, is a small molecule drug that inhibits B-cell proliferation and survival by irreversibly binding the protein Bruton's tyrosine kinase (BTK). Blocking BTK inhibits the B-cell receptor pathway, which is often aberrantly active in B cell cancers. Ibrutinib is therefore used to treat such cancers, including mantle cell lymphoma, chronic lymphocytic leukemia, and Waldenström's macroglobulinemia. Ibrutinib also binds to C-terminal Src Kinases. These are off-target receptors for the BTK inhibitor. Ibrutinib binds to these receptors and inhibits the kinase from promoting cell differentiation and growth. This leads to many different side effects like left atrial enlargement and atrial fibrillation during the treatment of Chronic Lymphocytic Leukemia. Jonathan Sessler
Jonathan Sessler (born 20 May 1956 in Urbana, Illinois) is a professor of chemistry at The University of Texas at Austin. He is notable for his pioneering work on expanded porphyrins and their applications to biology and medicine. He is a co-founder of Pharmacyclics, Inc., a company that works with expanded porphyrins, and Anionics, Inc., which develops anion recognition chemistry. Abexinostat (INN, formerly PCI-24781) is an experimental drug candidate for cancer treatment. It was developed by Pharmacyclics and licensed to Xynomic. and is in Phase II clinical trials for B-cell lymphoma.
