Open Field (animal Test)
Developed by Calvin S. Hall, the open field test is an experimental test used to assay general locomotor activity levels, anxiety, and willingness to explore in animals (usually rodents) in scientific research. However, the extent to which behavior in the open field measures anxiety is controversial. The open field test can be used to assess memory by evaluating the ability of the animal to recognize a stimuli or object. Another animal test that is used to assess memory using that same concept is the novel object recognition test. Concept Animals such as rats and mice display a natural aversion to brightly lit open areas. However, they also have a drive to explore a perceived threatening stimulus. Decreased levels of anxiety lead to increased exploratory behavior. Increased anxiety will result in less locomotion and a preference to stay close to the walls of the field (thigmotaxis). 5-HT1A Receptor
The serotonin 1A receptor (or 5-HT1A receptor) is a subtype of serotonin receptor, or 5-HT receptor, that binds serotonin, also known as 5-HT, a neurotransmitter. 5-HT1A is expressed in the brain, spleen, and neonatal kidney. It is a G protein-coupled receptor (GPCR), coupled to the Gi protein, and its activation in the brain mediates hyperpolarisation and reduction of firing rate of the postsynaptic neuron. In humans, the serotonin 1A receptor is encoded by the HTR1A gene. Distribution The 5-HT1A receptor is the most widespread of all the 5-HT receptors. In the central nervous system, 5-HT1A receptors exist in the cerebral cortex, hippocampus, septum, amygdala, and raphe nucleus in high densities, while low amounts also exist in the basal ganglia and thalamus. Tail Suspension Test
The tail suspension test (TST) is an experimental method used in scientific research to measure stress in rodents. It is based on the observation that if a mouse is subjected to short term inescapable stress then the mouse will become immobile. It is used to measure the effectiveness of antidepressant-like agents but there is significant controversy over its interpretation and usefulness. History The TST was introduced in 1985 due to the popularity of a similar test called the forced swim test (FST). However this test only recently became popular in the 2000s where data has shown that animals do show a change in behavior when injected with antidepressants. Hole-board Test
The hole-board test (HBT) is an experimental method used in scientific research to measure anxiety, stress, neophilia and emotionality in animals. Because of its ability to measure multiple behaviors it is a popular test in behavioral pharmacology, but the results are controversial. Concept The HBT was designed in the 1970s to mitigate the flaws of the open field test (OFT), another animal model that measures anxiety and exploratory behavior. Exploratory behavior requires a choice to be made to explore, but since the OFT is just simply a brightly lit area, the animal doesn’t have a choice. Forced Swim Test
The behavioural despair test (or Porsolt forced swimming test) is a test, centered on a rodent's response to the threat of drowning, whose result has been interpreted as measuring susceptibility to negative mood. It is commonly used to measure the effectiveness of antidepressants, although significant criticisms of its interpretation have been made. Method Animals are subjected to two trials during which they are forced to swim in an acrylic glass cylinder filled with water, and from which they cannot escape. The first trial lasts 15 minutes. Then, after 24-hours, a second trial is performed that lasts 5 minutes. The time that the test animal spends in the second trial without making any movements beyond those required to keep its head above water is measured. Light-dark Box Test
The light-dark box test (LDB) is a popular animal model used in pharmacology to assay unconditioned anxiety responses in rodents. The extent to which behavior in the LDB measures anxiety is controversial. Overview The LDB apparatus has two compartments. The light compartment is 2/3 of the box and is brightly lit and open. The dark compartment is 1/3 of the total box and is covered and dark. A door of 7 cm connects the two compartments. Rodents prefer darker areas over lighter areas. However, when presented in a novel environment, rodents have a tendency to explore. These two conflicting emotions lead to observable anxiety like symptoms. Rodents typically spend more time in the dark compartment than in the light compartment.
