Joshua Coon
Joshua Coon is a professor of chemistry and biomolecular chemistry and the inaugural holder of the Thomas and Margaret Pyle Chair at the University of Wisconsin–Madison, and an affiliate of the Morgridge Institute for Research. Coon develops scientific instruments to measure molecules in living systems. He focuses specifically on the use of mass spectrometry to measure the molecular mass of biomolecules, and its application to proteomics. Joshua Coon, along with John Syka, developed electron-transfer dissociation (ETD) while working as a postdoctoral student in Donald Hunt's lab at the University of Virginia. Early life and education Coon was born in Mount Pleasant, Michigan. Coon earned his B.Sc. from Central Michigan University in 1988. Mount Pleasant is a city in the U.S. state of Michigan. Located in Central Michigan, the city is the county seat of Isabella County. The population was 21,688 as of the 2020 United States census. It is surrounded by Union Township but is politically independent. Part of the city (with a population of 8,741) is located within the Isabella Indian Reservation, the base of the federally recognized Saginaw Chippewa Tribal Nation. The tribe's Soaring Eagle Casino & Resort in nearby Chippewa Township is also within the reservation boundaries. Electron-transfer Dissociation
Electron-transfer dissociation (ETD) is a method of fragmenting multiply-charged gaseous macromolecules in a mass spectrometer between the stages of tandem mass spectrometry (MS/MS). Similar to electron-capture dissociation, ETD induces fragmentation of large, multiply-charged cations by transferring electrons to them. ETD is used extensively with polymers and biological molecules such as proteins and peptides for sequence analysis. Transferring an electron causes peptide backbone cleavage into c- and z-ions while leaving labile post translational modifications (PTM) intact. The technique only works well for higher charge state peptide or polymer ions (z>2). However, relative to collision-induced dissociation (CID), ETD is advantageous for the fragmentation of longer peptides or even entire proteins. Mass is an intrinsic property of a body. It was traditionally believed to be related to the quantity of matter in a physical body, until the discovery of the atom and particle physics. It was found that different atoms and different elementary particles, theoretically with the same amount of matter, have nonetheless different masses. The Beckman Young Investigators Award was established by Mabel and Arnold Beckman in 1991, and is now administered by the Arnold and Mabel Beckman Foundation. Biemann Medal
The Biemann Medal is awarded annually by the American Society for Mass Spectrometry (ASMS) to an individual early in his or her career in recognition of significant achievement in basic or applied mass spectrometry. It is named after professor Klaus Biemann. Nominees must be within the first 15 years of receiving the Ph.D. in the year nomination is considered (exceptions may be considered). The Human Proteome Organization (HUPO) is an international consortium of national proteomics research associations, government researchers, academic institutions, and industry partners. Isobaric Labeling
Isobaric labeling is a mass spectrometry strategy used in quantitative proteomics. Peptides or proteins are labeled with chemical groups that have (at least nominally) identical mass (isobaric), but vary in terms of distribution of heavy isotopes in their structure. These tags, commonly referred to as tandem mass tags, are designed so that the mass tag is cleaved at a specific linker region upon high-energy CID (HCD) during tandem mass spectrometry yielding reporter ions of different masses. Quantitative Proteomics
Quantitative proteomics is an analytical chemistry technique for determining the amount of proteins in a sample. The methods for protein identification are identical to those used in general (i.e. qualitative) proteomics, but include quantification as an additional dimension. Rather than just providing lists of proteins identified in a certain sample, quantitative proteomics yields information about the physiological differences between two biological samples. For example, this approach can be used to compare samples from healthy and diseased patients.
