COS Cells
COS are fibroblast-like cell lines derived from monkey kidney tissue. COS cells are obtained by immortalizing CV-1 cells with a version of the SV40 virus that can produce large T antigen but has a defect in genomic replication. The CV-1 cell line in turn was derived from the kidney of the African green monkey. The acronym "COS" is derived from the cells being CV-1 (simian) in Origin, and carrying the SV40 genetic material. Three COS lines were created (COS-1, COS-3 and COS-7), of which two are commonly used (COS-1 and COS-7). Applications The COS cell lines are often used by biologists when studying the monkey virus SV40. Cells from these lines are also often transfected to produce recombinant proteins for molecular biology, biochemistry, and cell biology experiments. When an expression construct with an SV40 origin of replication is introduced into COS cells, the vector can be replicated substantially by the large T antigen The large tumor antigen (also called the large T-an ...

Fibroblast
A fibroblast is a type of cell (biology), biological cell that synthesizes the extracellular matrix and collagen, produces the structural framework (Stroma (tissue), stroma) for animal Tissue (biology), tissues, and plays a critical role in wound healing. Fibroblasts are the most common cells of connective tissue in animals. Structure Fibroblasts have a branched cytoplasm surrounding an elliptical, speckled cell nucleus, nucleus having two or more nucleoli. Active fibroblasts can be recognized by their abundant Endoplasmic reticulum#Rough endoplasmic reticulum, rough endoplasmic reticulum. Inactive fibroblasts (called fibrocytes) are smaller, spindle-shaped, and have a reduced amount of rough endoplasmic reticulum. Although disjointed and scattered when they have to cover a large space, fibroblasts, when crowded, often locally align in parallel clusters. Unlike the epithelial cells lining the body structures, fibroblasts do not form flat monolayers and are not restricted by a ...

Cell Line
An immortalised cell line is a population of cells from a multicellular organism which would normally not proliferate indefinitely but, due to mutation, have evaded normal cellular senescence and instead can keep undergoing division. The cells can therefore be grown for prolonged periods ''in vitro''. The mutations required for immortality can occur naturally or be intentionally induced for experimental purposes. Immortal cell lines are a very important tool for research into the biochemistry and cell biology of multicellular organisms. Immortalised cell lines have also found uses in biotechnology. An immortalised cell line should not be confused with stem cells, which can also divide indefinitely, but form a normal part of the development of a multicellular organism. Relation to natural biology and pathology There are various immortal cell lines. Some of them are normal cell lines (e.g. derived from stem cells). Other immortalised cell lines are the ''in vitro'' equivalent ...

SV40
SV40 is an abbreviation for simian vacuolating virus 40 or simian virus 40, a polyomavirus that is found in both monkeys and humans. Like other polyomaviruses, SV40 is a DNA virus that has the potential to cause tumors in animals, but most often persists as a latent infection. SV40 has been widely studied as a model eukaryotic virus, leading to many early discoveries in eukaryotic DNA replication and transcription. Human disease The hypothesis that SV40 might cause cancer in humans has been a particularly controversial area of research. It is currently unclear whether SV40 has any role in causing tumors. As a result of these uncertainties, academic opinion remains divided, with some arguing that this hypothesis is not supported by the data and others arguing that some cancers may involve SV40. The US National Cancer Institute announced in 2004 that although SV40 does cause cancer in some animal models, "substantial epidemiological evidence has accumulated to indicate that SV40 l ...

SV40 Large T-antigen
SV40 large T antigen ( Simian Vacuolating Virus 40 TAg) is a hexamer protein that is a dominant-acting oncoprotein derived from the polyomavirus SV40. TAg is capable of inducing malignant transformation of a variety of cell types. The transforming activity of TAg is due in large part to its perturbation of the retinoblastoma (pRb) and p53 tumor suppressor proteins. In addition, TAg binds to several other cellular factors, including the transcriptional co-activators p300 and CBP, which may contribute to its transformation function. TAg is a product of an early gene transcribed during viral infection by SV40, and is involved in viral genome replication and regulation of host cell cycle. SV40 is a double-stranded, circular DNA virus belonging to the Polyomaviridae (earlier Papovavirus) family, Orthopolyomavirus genus. Polyomaviruses infect a wide variety of vertebrates and cause solid tumours at multiple sites. SV40 was isolated by Sweet and Maurice Hilleman in 1960 in primary m ...

African Green Monkey
''Chlorocebus'' is a genus of medium-sized primates from the family of Old World monkeys. Six species are currently recognized, although some people classify them all as a single species with numerous subspecies. Either way, they make up the entirety of the genus ''Chlorocebus''. Confusingly, the terms "vervet monkey" and "green monkey" are sometimes used to refer to the whole genus ''Chlorocebus'', though they also refer more precisely to species ''Chlorocebus pygerythrus'' and '' Chlorocebus sabaeus'', respectively, neither of which is the type species for ''Chlorocebus''. This article uses the term ''Chlorocebus'' consistently for the genus and the common names only for the species. The native range of these monkeys is sub-Saharan Africa from Senegal and Ethiopia south to South Africa. However, in previous centuries, a number of them were taken as pets by early Caribbean settlers and slave traders, and were transported across the Atlantic Ocean to the Caribbean islands. Th ...
