Andrea Ballabio
Andrea Ballabio (born in Naples, Italy, January 27, 1957) is an Italian scientist and academic professor. He is director of the Telethon Institute of Genetics and Medicine (TIGEM) of Pozzuoli; professor of medical genetics at the Federico II University of Naples and visiting professor of genetics at Baylor College of Medicine in Houston, Texas, US and at the University of Oxford-UK. Early life and education After graduating in medicine and specializing in pediatrics at the Federico II University in Naples, he spent many years abroad, first in England and then in the United States, where he became associate professor at the Department of Molecular and Human Genetics, Baylor College of Medicine and co-director of Baylor Human Genome Center in Houston, Texas. TFEB
Transcription factor EB is a protein that in humans is encoded by the ''TFEB'' gene. Function TFEB is a master gene for lysosomal biogenesis. It encodes a transcription factor that coordinates expression of lysosomal hydrolases, membrane proteins and genes involved in autophagy. Upon nutrient depletion and under aberrant lysosomal storage conditions such as in lysosomal storage diseases, TFEB translocates from the cytoplasm to the nucleus, resulting in the activation of its target genes. TFEB overexpression in cultured cells induces lysosomal biogenesis, exocytosis and autophagy. In bacterial infection nicotinic acid adenine dinucleotide phosphate (NAADP) induction of lysosomal Ca2+ efflux and TFEB activation leads to enhanced expression of inflammatory cytokines. The European Research Council (ERC) is a public body for funding of scientific and technological research conducted within the European Union (EU). Spinal muscular atrophy (SMA) is a rare neuromuscular disorder that results in the loss of motor neurons and progressive muscle wasting. It is usually diagnosed in infancy or early childhood and if left untreated it is the most common genetic cause of infant death. It may also appear later in life and then have a milder course of the disease. The common feature is progressive weakness of voluntary muscles, with arm, leg and respiratory muscles being affected first. Associated problems may include poor head control, difficulties swallowing, scoliosis, and joint contractures. Alpha-1 antitrypsin deficiency (A1AD or AATD) is a genetic disorder that may result in lung disease or liver disease. Onset of lung problems is typically between 20 and 50 years of age. This may result in shortness of breath, wheezing, or an increased risk of lung infections. Complications may include chronic obstructive pulmonary disease (COPD), cirrhosis, neonatal jaundice, or panniculitis. A1AD is due to a mutation in the SERPINA1 gene that results in not enough alpha-1 antitrypsin (A1AT). Risk factors for lung disease include tobacco smoking and environmental dust. The underlying mechanism involves unblocked neutrophil elastase and buildup of abnormal A1AT in the liver.
